BPC-157 Guide: What It Is, Research Benefits and Side Effects

What Is BPC-157?

BPC-157 (Body Protection Compound-157) is a synthetic pentadecapeptide composed of 15 amino acids with the sequence Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val. It was isolated and characterised from a larger protein fraction present in gastric juice, and its molecular formula is C62H106N16O22, with a molecular weight of approximately 1,419.53 Da.

The compound is water-soluble and demonstrates notable stability under physiological pH conditions, which has made it a subject of interest in biochemistry and molecular biology research settings.

Structural Characteristics

The peptide chain of BPC-157 features a high concentration of proline residues at positions 3, 4, 5, and 8. This proline-rich region is thought to confer conformational rigidity and resistance to proteolytic degradation — properties relevant to its behaviour in aqueous research assays. The presence of glutamic acid and aspartic acid residues contributes to the overall negative charge of the molecule at physiological pH.

X-ray crystallography and NMR studies have characterised the peptide as adopting a relatively extended conformation, though secondary structure formation is influenced by solvent conditions and ionic concentration in the assay environment.

Receptor and Signalling Interactions of Interest

Research investigations have explored BPC-157’s interactions with several molecular pathways:

  • Nitric Oxide (NO) Pathway: In-vitro studies have observed modulation of endothelial nitric oxide synthase (eNOS) activity in cell culture models, suggesting potential interactions with the NO signalling cascade relevant to vascular biology research.
  • Growth Factor Receptor Modulation: Laboratory data indicates that BPC-157 may influence expression levels of VEGF (vascular endothelial growth factor) receptors in fibroblast and endothelial cell lines, making it a compound of interest for angiogenesis research.
  • FAK–Paxillin Pathway: Cell migration assays have recorded activation of focal adhesion kinase (FAK) and its downstream effector paxillin in the presence of BPC-157, a pathway implicated in cytoskeletal reorganisation and cellular motility studies.
  • Dopaminergic System: Rodent model studies have investigated BPC-157’s effect on dopamine turnover in limbic structures, with some data pointing to interactions with dopamine D1 and D2 receptor expression in striatal tissue.

Stability and Handling in Research Conditions

BPC-157 demonstrates good aqueous solubility (recommended: sterile water or phosphate-buffered saline, pH 7.4) and is stable at −20 °C when lyophilised. Repeated freeze-thaw cycles are documented to reduce bioactivity in cell-based assays; working aliquots should be prepared accordingly. The peptide is not recommended for storage in solutions above 37 °C for extended periods.

HPLC analysis of research-grade BPC-157 should confirm purity ≥98%, with mass spectrometry (ESI-MS or MALDI-TOF) used to verify the expected [M+H]+ ion at m/z 1420.6.

Research Applications

BPC-157 is used as a research tool across several pre-clinical disciplines:

  • Angiogenesis and wound biology in cell culture and rodent models
  • Gastrointestinal mucosal biology — the compound’s origin from gastric juice proteins gives it relevance in gastric epithelial cell research
  • Tendon and ligament cell (tenocyte) proliferation assays
  • Neurochemical studies examining monoamine system interactions in animal models

All use of BPC-157 is confined to authorised laboratory settings. It is supplied by PeptidesNextDay as a reference-grade research compound, accompanied by a full certificate of analysis.

Certificate of Analysis

Each batch of BPC-157 supplied by PeptidesNextDay is independently verified by a UKAS-accredited third-party laboratory. Testing includes reverse-phase HPLC purity assessment, ESI mass spectrometry for identity confirmation, and endotoxin testing where applicable. A full CoA is available for download from the product page.

BPC-157 benefits reported in research

Almost all BPC-157 data come from animal models, chiefly rats. Within that limitation, the studies report a consistent set of findings that explain why it is one of the most searched research peptides in the UK:

  • Gastrointestinal protection. Reduced lesion size in models of gastric ulcer, NSAID-induced injury, inflammatory bowel disease and fistula.
  • Tendon, ligament and muscle repair. Faster functional recovery in rat Achilles tendon transection, medial collateral ligament injury and crush-injury models, with increased fibroblast migration and collagen organisation.
  • Angiogenesis. Up-regulation of VEGFR2 and increased new vessel formation in ischaemic tissue.
  • Nitric-oxide system modulation. Counteraction of both NOS inhibition and NO over-production in several models, proposed as a common pathway for its effects.
  • Neuro-protection and dopamine-system effects in a smaller number of rodent studies.

No adequately powered, peer-reviewed human trial of BPC-157 has been published. Claims of “benefits” in people are extrapolations from animal data.

BPC-157 side effects

In the animal literature BPC-157 has been reported as well tolerated, with no lethal dose identified in the toxicity studies that have been published and no consistent organ toxicity described. That is not the same as a demonstrated human safety profile: there are no controlled human safety data, no pharmacovigilance record and no established contraindications. Theoretical concerns raised in reviews include its pro-angiogenic activity (relevant to any condition where new blood-vessel growth is undesirable) and unknown interactions. In 2022 WADA added BPC-157 to the Prohibited List under S0 (non-approved substances), and in 2023 the US FDA placed it in Category 2 of its bulk drug substances list, citing safety concerns and restricting compounding. Anecdotal reports online describe nausea, dizziness or injection-site reactions, but these are unverified.

Oral BPC-157 vs injectable

A distinctive feature of BPC-157 in the literature is that it is reported to be stable in gastric juice and active when given orally in rodents, which is where interest in “oral BPC 157” and BPC-157 capsules comes from. In research settings the lyophilised peptide is reconstituted for parenteral models or dissolved for oral-gavage models. Capsule formulations sold commercially have not been tested in peer-reviewed studies. PeptidesNextDay supplies BPC-157 5mg as a lyophilised research-grade powder with batch COA; we do not currently stock a capsule format.

BPC-157 and TB-500 together

BPC-157 is frequently studied and discussed alongside TB-500 (Thymosin Beta 4), sometimes informally called a “Wolverine” blend. The two peptides have different mechanisms — BPC-157 acting largely through growth-factor and NO pathways, TB-500 through actin sequestration and cell migration — which is why they are combined in tissue-repair research. Read our TB-500 guide for the actin-binding side of that pairing, and use the peptide calculator to work out concentrations for a blended vial.

BPC-157 is not a controlled substance under the Misuse of Drugs Act and is not a licensed medicine. It may be lawfully sold for laboratory research; it may not be marketed or supplied for human consumption. It is prohibited in sport under WADA rules.
